Check www.voodoofiles.com for your card drivers and www.glsetup.com for your opengl driver for your card.

For the "software mouse under the normal mouse" problem:
I recently played another game (not Uplink), where I had the same error (two mouse cursors). I discovered by chance that when I switched to the windows desktop and back, the hardware mouse cursor vanished, and only the software mouse cursor remained.
So, you might try the following: Hit Alt+Tab to switch to the windows desktop, then hit Alt+Tab again (might have to hold down Alt and hit Tab several times to choose the correct program (Uplink)) to switch back to Uplink.
I don't know if that helps, never tried that with Uplink...
