Do they need to be staff only so inmates are escorted by guards or set to everyone!?
At first I thought they should be set to staff only so only guards could open it but realized a lot of them were wanting to go back to the cells to sleep during freetime. So no it's not necessary. Although I wonder if you had like an equal amount of guards and prisoners would everyone get the escort to the cell that they'd need?

No, prisoners are only escorted by guards when they're being put into a cell or taken to solitary/medical ward.
No don't lock the cell doors - the inmates need to be able to walk to the showers, kitchen, yard and common room on their own. The game doesn't require the typical "chain gang" principle where guards escort people out.
Your guards will escort inmates past the staff only doors when required (visit to medbay, shortcuts between buildings only for staff et cetera)
The only places you'll want to set staff only are kitchen, office and storage areas to keep the prisoners from stealing makeshift weapons like knives. You'll want metal detectors as well to inform guards of hidden weapons
